阿托伐他汀对实验性变态反应性脑脊髓炎外周血单个核细胞免疫功能的影响(1)

     【摘要】 目的 观察阿托伐他汀对豚鼠实验性变态反应性脑脊髓炎(EAE)外周血单个核细胞分泌细胞因子(CK)能力的影响,探讨其防治EAE的免疫调节机制。方法 采用粗制碱性髓鞘蛋白加福氏完全佐剂皮下注射的方法建立EAE模型。将40只豚鼠随机分成4组:正常对照组、EAE对照组、EAE低剂量治疗组和EAE高剂量治疗组,每组10只,造模前5d正常对照组和EAE对照组每天灌喂氯化钠溶液,EAE低剂量、高剂量治疗组分别在造模前5d开始灌喂含阿托伐他汀1mg/(kg•d)、4mg/(kg•d),直至整个实验结束。酶联免疫吸附法测定各组发病高峰期外周血单个核细胞分泌的细胞因子IFN-γ、IL-4、IL-10、TNF-α的水平。结果 EAE对照组外周血单个核细胞分泌IL-4、IL-10的能力较正常对照组明显低(P<0.05),EAE高、低剂量治疗组外周血单个核细胞分泌IL-4、IL-10的能力较EAE对照组明显增强(P<0.05)。EAE对照组外周血单个核细胞分泌IFN-γ、TNF-α的能力较正常对照组明显增强(P<0.05),EAE高、低剂量治疗组外周血单个核细胞分泌IFN-γ、TNF-α的能力较EAE对照组明显减弱(P<0.05)。结论 阿托伐他汀防治EAE的免疫机制可能是通过抑制IFN-γ、TNF-α产生及促进IL-4、IL-10产生而发挥。

    Study on Variation of Peripheral Blood mononuclear Cellular Immune of Guinea pig with EAE after Treating with Atorvastatin

    【Abstract】 Objective To explore the mononuclear Cellular immune of experimemtal allergic encephalomyelitis and the immunol regulating mechanisms of Atorvastatin on EAE. Methods Induced experimemtal allergic encephalomyelitis in Guinea pig by subcutaneous administrating coarse myelin basic protein. 40 Guinea pigs were assigned randomly to 4 groups: the normal group, the EAE control group, the Atorvastatin low dose treating EAE group and the Atorvastatin high dose treating EAE group.The normal group and EAE control group were perfused with the water and low dose、high dose treating EAE group were perfused with Atorvastatin 1 mg/(kg•d)、4 mg/(kg•d) separately from 5 days before Modeling. The levels of cytokine IL-4、IL-10、IFN-r and TNF-αin peripheral blood mononuclear cells were detected at fastigium by use of ELISA. Results The levels of IL-4、IL-10 in peripheral blood mononuclear cells in EAE control group were lower than those in the normal group(P<0.05). Comparing to EAE control group,low dose treating EAE group group or high dose treating EAE group increased (P<0.05).While in EAE control group, the levels of IFN-γ、TNF-αin peripheral blood mononuclear cells were higher than those in the normal group (P<0.05).
