玉竹对UVB诱导HaCaT细胞损伤的保护作用(1)

     [摘要]目的:探讨玉竹水提液对中波紫外线(UVB)诱导人皮肤角质形成细胞损伤的保护作用及其可能机制。方法:用64mJ·cm-2的UVB照射人皮肤角质形成细胞建立光老化细胞模型,以不同浓度玉竹提取物处理光老化细胞,羟胺法检测细胞超氧化物歧化酶(SOD)活性,比色法检测谷胱甘肽过氧化物酶(GSH-Px)活性,TBA法检测丙二醛(MDA)含量,酶联免疫法检测肿瘤坏死因子α(TNF-α)、白介素6(IL-6)分泌量。结果:UVB照射角质形成细胞后,SOD、GSH-Px活性降低,MDA含量升高,TNF-α、IL-6分泌量增加(P<0.01),玉竹水提液中(100μg·ml-1)、高(200μg·ml-1)剂量组能显著提高SOD、GSH-Px活性,降低MDA含量,减少TNF-α、IL-6分泌(P<0.05或P<0.01)。结论:玉竹水提液可在一定程度上减轻UVB对角质形成细胞的损伤,其机制可能与其抑制氧化损伤,增强细胞抗氧化能力,减少炎症细胞因子分泌有关。

    Protective effects of yuzhu on hacat cells damaged by ultraviolet B

    Abstract: Objective To investigate the protective effect of water-extract solution of Yuzhu on HaCaT cells injured by irradiation of ultraviolet B and its possible mechanisms. Methods Cell photoaging model was established by irradiating HaCaT cells with UVB(64mJ·cm-2). HaCaT cells were treated with different concentration water-extract solution of Yuzhu. The activity of superoxide dismutase (SOD) and glutathione peroxidase(GSH-Px),the content of malondialdehyde(MDA) were detected by biochemical assay, the secretion levels of TNF-α and IL-6 were detected by enzyme-linked immunosorbent assay (ELISA). Results The proliferation activity of HaCaT cells descended with the UVB irradiation, the activities of SOD and GSH-Px decreased, when the content of MDA, TNF-α, IL-6, were ascensive(P<0.01). But under the intervention at middle concentration(100μg·ml-1) and high concentration(200μg·ml-1) of water-extract solution of Yuzhu, the above changes induced by UVB irradiation were inhibited(P<0.05 or P<0.01). Conclusion The water-extract solution of Yuzhu might inhibit the proliferation decreasing which induced by UVB. And its mechanism may have relationship with the ability of strengthening antioxidation, decreasing oxygen radicals and decreasing the secretions of proinflammatory cytokines ......
